Adhering suspended samples onto coverslips

Solution: 0.1% polyethyleneimine

  • 50% polyethyleneimine, aqueous (Sigma P-3143): 20.0 µL
  • Q.S. with distilled H2O: 10.0 mL

Procedure

  1. Clean coverslips with 95% EtOH and allow to air dry

  2. Add drop of 0.1% polyethyleneimine to coverslip and spread drop to cover surface of coverslip. Allow to incubate for 1-2 minutes. Do not allow to dry.

  3. Briefly rinse coverslip with distilled water and allow to air dry (usually 10-20 minutes, use within 1 hour).

  4. Add sample suspension to coverslip and allow to adhere for 10-15 minutes.

  5. Wick off excess solution with filter paper and wait additional 2-3 minutes. DO NOT ALLOW TO AIR DRY!

  6. Specimens are now mounted and ready to continue with additional processing.
